The basement membrane, also known as base membrane, is a thin, pliable sheet like type of extracellular matrix that provides cell and tissue support and acts as a platform for complex signalling. The basement membrane sits between epithelial tissues including mesothelium and endothelium, and the underlying connective tissue. The underlying connective tissue attaches to the basal lamina with collagen VII anchoring fibrils and fibrillin microfibrils. The basal lamina layer can further be subdivided into two layers based on their visual appearance in electron microscopy. The lighter colored layer closer to the epithelium is called the lamina lucida, while the denser colored layer closer to the connective tissue is called the lamina densa. The electron dense lamina densa layer is about 30–70 nanometers thick and consists of an underlying network of reticular collagen IV fibrils which average 30 nanometers in diameter and 0.1–2 micrometers in thickness and are coated with the heparan sulfate rich proteoglycan perlecan. In addition to collagen, this supportive matrix contains intrinsic macromolecular components. The lamina lucida layer is made up of laminin, integrins, entactins, and dystroglycans. Integrins are a key component of hemidesmosomes which serve to anchor the epithelium to the underlying basement membrane. Основная функция базальной мембраны заключается в закреплении эпителия к расположенной под ним рыхлой соединительной ткани (дерме или собственной пластинке слизистой). Это достигается посредством адгезии клеток к матриксу через молекулы адгезии к субстрату (SAM). Базальная мембрана действует как механический барьер, предотвращая инвазию злокачественных клеток в более глубокие ткани. Ранние стадии злокачественного процесса, ограниченные эпителиальным слоем базальной мембраной, называются карциномой in situ. Базальная мембрана также необходима для ангиогенеза (развития новых кровеносных сосудов). Белки базальной мембраны способствуют ускорению дифференцировки эндотелиальных клеток. Наиболее известными примерами базальных мембран являются гломерулярная базальная мембрана почки, формирующаяся в результате слияния базальной ламины эндотелия гломерулярных капилляров и базальной ламины подоцитов, а также базальная мембрана между альвеолами легких и легочными капиллярами, образующаяся при слиянии базальной ламины альвеол и базальной ламины капилляров легких, где происходит диффузия кислорода и газов (газообмен). По состоянию на 2017 год, к другим функциям базальной мембраны относятся фильтрация крови и поддержание мышечного гомеостаза.
Эволюционное происхождение
Они встречаются только у диплобластных и гомосклероморфных губок. В некоторых исследованиях гомосклероморфы оказались сестринской группой для диплобластов, что указывает на единое происхождение этой структуры в истории жизни. Однако более поздние исследования отвергли объединение диплобластов и гомосклероморф, поэтому, возможно, другие губки утратили её (наиболее вероятно), или же она возникла независимо в обеих группах.
